How to reconstitute a lyophilized peptide vial
- 1Bring the vial to room temperatureLet the lyophilized vial equilibrate for 15–20 minutes so it isn't cold when the solvent contacts the cake.
- 2Measure the solventDraw the volume of sterile solvent you entered above (bacteriostatic water is typical, commonly 1–3 mL) with a graduated syringe or pipette.
- 3Add slowly along the side wallRun the solvent slowly down the inside wall of the vial. Never jet it directly onto the lyophilized cake.
- 4Swirl — do not shakeGently swirl until the cake fully dissolves. Shaking shears fragile peptide sequences and can denature them.
- 5Store refrigerated and record the concentrationKeep the reconstituted vial at 2–8 °C and label it with its mg/mL concentration and the date of reconstitution.
